Izak Colthof is op 26 april 1916 met Jenny Hammelburg getrouwd in Nijkerk. Uit dit huwelijk zijn 8 kinderen geboren. Het eerste kindje, Esther, is op 5-jarige leeftijd overleden, en de andere 7 kinderen zijn allen omgekomen.
