Israel Colthof was een zoon van Aaron Colthof en Ester Hirsch. In 1936 trouwde hij met Marianne Frank. Op 18 november 1939 werd hun eerste zoon geboren, Aaron Sjoloum Colthof. Het jongetje is maar negen dagen oud geworden. Hij overleed op 27 november 1939.
